Which version of MonoDevelop are you using? The one linked from the
http://monodevelop.com/Download/Mac_MonoTouch page?

Can you confirm that the directory /Developer/MonoTouch and the file
/Developer/MonoTouch/usr/lib/mono/2.1/mscorlib.dll exist?
