My GTK# assemblies are in 

C:\Program Files\Common Files\GTK\2.2\lib\gtk-sharp

So should yours be...

This command should work:

mcs -lib:"C:\Program Files\Common Files\GTK\2.2\lib\gtk-sharp"
-r:gtk-sharp -r:System.Drawing gui.cs

All library assemblies are .dll files in the .NET/Mono world.
